Protein Engineering and Structural biology Lab Sanquin research, Amsterdam |
The goal is to develop a novel protein tagging system for fast, cheap and facile protein purification. Based on recent in silico design work in our lab resulting in lead peptide candidates against a target protein, we would like to optimize binding (affinity maturation) of these by modifying both peptide and target protein simultaneously using directed co-evolution by compartmentalized two-hybrid replication. Resulting peptide-protein pairs will be tested by co-immunoprecipitation and affinity determined by surface plasmon resonance. The best protein variants will be conjugated to beads for affinity purification of proteins expressing the respective peptide tag. |
